The two phases are an aqueous (water-based) solvent and an organic solvent. A reaction was conducted in one of these solvents. The product of this reaction will be more soluble in the other solvent, which was added to the reaction mixture to extract the product. As water and organic solvents do not mix (they are immiscible), the mixture has separated out into the two layers (clear, yellow) in the funnel. The tap (red) is turned to obtain the layer with the product in it.
